Abstract (EN)

Bacteriophages representing the most abundant biological community in nature are defined as viruses that infect bacteria. One of the most well-known properties of bacteriophages is that they are species specific. Due to the crises in the production of new antibiotics and the increasing use of antibiotics around the world, a search for an alternative approach to antibiotic treatment against resistant bacteria has come. In parallel with this, the therapeutic use of bacteriophages (Phage Therapy), which had actually existed for 100 years, has again become the focus of attention in the scientific and medical world today. Pathogenic Escherichia coli show by World Health Organization (WHO) as one of the most important factors of infectious diseases that are difficult to treat in hospital and community. Within the scope of this thesis; Isolation of lytic bacteriophages effective against E. coli DH10B strain from environmental samples and determination of the activity of these phages against Escherichia coli isolates originating from diabetic foot infection (DAI) were aimed. In the study, 15 bacteriophages with different plaque morphologies were isolated and restriction endonuclease profiles were determined. It is determine that there are 4 different cut profiles among the 9 phages that were cut, and 3 different phage biotypes were determined according to their lytic activity on E. coli strains among the 6 uncut phages. Phages with different profiles from KK1 to KK7 were coded and their efficacy against the 30 clinical isolates included in the study was tested. It was determined that KK4 phage with the highest lytic activity was effective on 22 (73%) of the isolates and the cocktail prepared from the mixture of all phages affected all bacteria except 3 isolates (90%). Of phages, including the phages we isolated, have the potential to be used with antibiotics for infections that do not respond to treatment. Especially KK4 phage, which is found to be effective on 73% of clinical E. coli isolates, has a high potential to be used for this purpose.
