Abstract (EN)
In this study, Clostriudum sp. The amylase enzyme was produced and the temperature, pH and substrate concentration were determined on the enzyme activity and the effect of the produced enzyme on biogas production in starch-containing wastewater was determined. The reproductive temperature of the bacteria isolated in the study is 37 ° C and is mesophilic. The effect of temperature on enzymatic activity has been determined between 30-80C and pH 'between 3.0-8.0. The optimum temperature at which the enzyme works was determined as 30C and pH 6.0. In the enzymatic hydrolysis study, which was performed depending on time, the rate of indirect sugar released after 8 hours is 3.65 mg / mL, which is 4.38 times higher than the rate of sugar released in one hour. The effect of the substrate concentration was determined at 0.5, 0.75, 1 and 2 starch concentrations, and when the substrate concentration doubled, the reducing sugar content increased by 39%. The effect of using enzyme on biogas production was determined at 37ºC and 3400 mL biogas was produced in the control group without enzyme, whereas 3670 mL 55% CH4 and 44% CO2 gas were produced in the experimental group. It has been determined that the addition of enzyme provides 7.3% more biogas production. Biogas production reached equilibrium 4 days earlier than the control group. In this study, anaerob Clostriudum sp. It can be said that the amylase isolated from bacteria can be used in biogas systems. Keywords: Clostriudum sp., Biogas, Amylase, Enzyme Activity
