New perspective of childhood astim pathogenesis and clinic: E-cadherin gene promoter region methylation

Abstract (EN)
Aim: Our purpose in this study, ın patients with asthma, which is the promoter of the epithelial barrier of E-cadherin protein molecule to evaluate the methylation r, to understand their role in asthma pathogenesis of epithelial barrier and the consequent epithelial protein is to contribute to the development of current treatment modalities in the treatment of asthma. Material methods: Asthmatic patients between 5-15 years of age will be included in the study and this paitent more than 3 months ago with no history of inhaled or systemic corticosteroids. Patients admitted to Celal Bayar University Hospital Pediatric Emergency Service or Pediatric Clinic will be included in this study. 20 atopic asthma, 20 non-atopic asthma, 10 control group will be a total of 50 cases . Age, gender information, a family history of atopy, routine tests, skin prick tests, pulmonary function tests, total IgE levels will be saved. When Routine blood tests taken during the patient's peripheral blood , 2 cc of peripheral blood to be separated for DNA isolation . The promoter of ECAD gene will be evaluated methylation. İn this DNA sample to be applied methylation specific PCR. Results: 20 atopic asthma, 22 nonatopic asthma, and 20 control groups were evaluated for e-cadherin promoter region methylation in the study. When the methylation status of the subjects included in the study were evaluated according to the groups, no statistically significant difference was found among the individuals in the different groups (p= 0,734). The methylation status of individuals in different study groups is similar. Conclusion: All studies show that epigenetic modifications and E-cadherin have an important role in the etiopathogenesis of asthma. To investigate the effect of epigenetic factors on E-cadherin for this purpose, we looked at the rates of methylation in the E-cadherin promoter region in asthmatic patients and in the control group. There were no statistically significant differences between the groups when the groups included in the study included 62 subjects who were included in the study.
