High incidence of tuberculosis in developed and developing countries and apperaring of strains with resistance to two major drugs (MDR) in addition to two minor drugs (XDR) and tendency to spread of beijing strains which does not give response to anti-tuberculosis treatment in the whole world are the most important handicaps of tuberculosis control programmes. This global problem can be exceed by monitoring M. Tuberculosis strains with more sensitive and more discriminative molecular epidemiologic methods. In this manner molecular methods and modifications like PCR, PCR-RFLP, MIRU-VNTR and spoligotyping which targets polymorphism of short repeats distrubiting randomly in genome, Hsp65 and IS6110 regions had been tested. In such studies made, it is concluded that the discriminative power of these methods is lower if they are used lonely in contrast to combined use. In our previous studies some of these methods have been tested in our labarotories to determine the epidemiological properties of M. Tuberculosis strains isolated from pulmonar tuberculosis patients and presence of Beijin strain had been demonstrated. In this study the possible contribution to epidemiologic studies of high discriminative power which will be obtained by combining 3 methods will be concluded in addition to questioning of relations between M. Tuberculosis strains with IS6110-RFLP, 15 locus MMIRU-VNTR and spoligotyping. Within this project; the sputum samples which sent to C.U.T.H.A.U.M. Regional Tuberculosis Lab. And grown on liquid and solid culture media and diagnosed as M. Tuberculosiscomplex members with commercial card tests will be examined for phylogenetic relations and will be diagnosed in species level with IS6110, MIRU-VNTR and spoligotyping methods.
