The effect of different growth regulators, explant types and genotype on organogenesis in C.coum and C.persicum

In this thesis, regeneration by organogenesis technique was searched using leave, petiole and rhizome explants of C. coum and C. persicum species. Different combination and concentrations of 7 different plant growth regulators (GA3, Prolin, Spermin, NAA, IBA, BA ve Kinetin) and MS,B5, ROM ve WPM media were tested to get callus and shoot structures in C.coum ve C.persicum species. Callus and shoot formation were observed from the media with GA3, Prolin, Spermin, NAA and Kinetin. Nothing was observed from the media with IBA ve BA. The highest regeneration ratio was obtained from the experiments coming from in vitro C.persicum plants and C.coum germinated from seeds. In this experiment, callus formation ratios were obtained from petiole, leave and rhizome explants as 15.6%, 13.5% and 23.9%. In C.persicum, the best interaction between media and explant was observed from MS medium and rhizome explants (39.6%) for callus formation, WPM medium and petiole (14.58 %) for shoot formation, WPM medium and petiole (12.5 %) for root formation. In C.coum, callus formation ratios were obtained from petiole, leave and rhizome explants as 20.8%, 12.5% and 32.3%. In C.coum, the best interaction between media and explant was observed from WPM medium and rhizome explants (45.8 %) for callus formation, B5 medium and rhizome (16.6 %) for shoot formation, ROM medium and leave explant (12.5 %) for root formation. Callus formation was observed from 1/2 MS media with 0.5 mg/l GA3, 1 mg/l prolin, 0.05 mg/l spermin, 3 mg/l Kinetin ve 1 mg/l NAA in both C.persicum and C. coum species.
