Investigation of protective effects of naringin on EKG and cardiac enzymes in cyclophosphamide-induced rats

Abstract (EN)

Aim: Cyclophosphamide (CYP) is an antineoplastic chemotherapeutic agent and a widely used anticancer and immunosuppressant drug. Recently, the number of studies using plant-based compounds has been increasing in order to minimize the cytotoxic effects of antineoplastic drugs. One of the compounds used for this purpose is Naringin. In this study, it is aimed to investigate the protective effects of Naringin on EKG and heart enzymes in CYP-induced rats. Material and Methods: Forty male adult Sprague Dawley rats weighing approximately 200-250 g were used in our study. Eight rats were used in each group and 5 groups were formed. All animals were subjected to standard maintenance and feeding conditions. Group 1 received saline (1 ml) intragastric (i.g.) for 10 days. Group 2 received 10 days intragastric serum physiologic was given and intraperitoneal (i.p.) CYP (200 mg / kg) was given at the 10th day in a single dose. Groups 3 and 4 were treated for 10 days with 50 and 100 mg / kg, respectively, in saline solubilized Naringin was administered and a single dose of CYP (200 mg / kg, i.p.) was administered after 10 days of naringin injection. Group 5 received 100 mg / kg i.g. solubilized in saline only for 10 days Naringine was applied. Forty-eight hours after the CYP administration, on the 12th day of the experiment, after EKG recorded intracardiac blood samples were taken under anesthesia with rats thiopental sodium (20 mg / kg) and sacrificed by cervical dislocation method. Analyzes of heart enzymes were performed in blood and serum samples obtained. To troponin I, Rat Cardiac Troponin Test Kit (Cat. No: KT-478) was screened by ELISA. CK and CK-MB were assigned to the Modular pp auto analyzer. Results: Cyclophosphamide has been shown to have cardiotoxic effects on ECG values and cardiac enzymes of rats. The number of heart beats(minute) in the groups were 375 ± 30 in the control group, 510 ± 17 in the CYP group, 494 ± 28 in the Naringin50 + CYP group, 475 ± 27 in the Naringin 100 + CYP group and 385 ± 26 in the Naringin 100 group. Conclusion: It has been found that different doses of naringin have the inhibiting cardiotoxicity effects of cyclophosphamide induced rats on ECG values and cardiac enzymes
