Abstract (EN)

Protective Effects of Naringin on Liver Enzymes (AST, ALT, ALP) in Cyclophosphamide-Induced Rats Aim: Cyclophosphamide (CYP) is an antineoplastic chemotherapeutic agent and a widely used anticancer and immunosuppressant drug. Antineoplastic chemotherapeutic agents generally have done cytotoxic effects on one or more target tissues. Recently, the number of studies using plant-based compounds has been increasing in order to minimize the cytotoxic effects of antineoplastic drugs. One of the compounds used for this purpose is Naringin. Naringine is a vegetable flanovoid. In this study, it is aimed to investigate the protective effects of Naringin on liver enzymes (AST, ALT, ALP) in CYP-induced rats. Material and Methods: Forty male adult Sprague Dawley rats weighing approximately 200-250 g were used in our study. Eight rats were used in each group and 5 groups were formed. All animals were subjected to standard maintenance and feeding conditions. Group 1 received saline (1 ml) intragastric (i.g.) for 10 days. Group 2 received 10 days intragastric serum physiologic was given and intraperitoneal (i.p.) CYP (200 mg / kg) was given at the 10th day in a single dose. Groups 3 and 4 were treated for 10 days with 50 and 100 mg / kg, respectively, in saline solubilized Naringin was administered and a single dose of CYP (200 mg / kg, i.p.) was administered after 10 days of naringin injection. Group 5 received 100 mg / kg i.g. solubilized in saline only for 10 days Naringine was applied. Forty-eight hours after the CYP administration, on the 12th day of the experiment, intracardiac blood samples were taken under anesthesia with rats thiopental sodium (20 mg / kg) and sacrificed by cervical dislocation method. Blood samples from rats were transferred to gli tubes with clot factor and centrifuged at 3500-4000 rpm for 10 minutes in a refrigerated centrifuge at +4 °C. The resulting serum samples were aliquoted and stored at -80 ° C in freezer until analysis. Analyzes of liver enzymes were done in auto analyzer and spectrophotomete. The quantitative values obtained at the end of the studies were evaluated by applying the one-way ANOVA Duncan test used in statistical analysis of more than two independent groups in SPSS 20.00 statistical data program. Results: CYP was found to cause hapatotoxicity and showed increased liver enzymes in rats. AST, ALT and ALP values were 58 ± 11, 32 ± 8, 38 ± 6 in the control group; 237 ± 42, 168 ± 44, 74 ± 11 in the CYP group; 223 ± 33, 158 ± 42, 62 ± 9 in the Naringin 50 + CYP group; 117 ± 25, 07 ± 24, 48 ± 8 in naringin 100 + CYP group; 54 ± 9, 31 ± 7, 36 ± 6 in the group of naringin 100 were found respectively. According to the values obtained from the control group, there is an increase of p <0.001 in all of the AST, ALT and ALP values in CYP-treated groups. Some of the values in the 2 groups given naringin 50 + CYP and Naringin 100 + CYP compared to values of only CYP groups were reduced significantly statistical (p<0.05). Conclusion: Different doses of naringin were detected to prevent of CYP adverse effects and suppression of hepatotoxicity. Keywords: : CYP, Liver Enzeymes (AST, ALT, ALP) , Naringin, Rat.
