Comparison of high performance liquid chromatography, mass spectrometry and chemiluminescence methods for vitamin D measurement

Abstract (EN)
The levels of 25(OH) vitamin D is measured as a clinical marker to determine the level of vitamin D in the body. Therefore, it is important to measure and to standardize the levels of 25(OH) vitamin D. In the present study, it was aimed to measure the levels of 25(OH) vitamin D by High Performance Liquid Chromatography (HPLC), mass spectrometry and chemiluminescence methods and to compare the results obtained in three ways. A total of 57 volunteers, including 14 males (23.7%) and 45 females (76.3%), included to the study. As a result of measurements made by three different methods, 25(OH) vitamin D levels were measured as 19.73 ± 11.87 ng / ml (approx. ± SD) by chemiluminescence method, 18.87 ± 12.47 ng / mL by HPLC method and 18.40 ± 10.90 ng / ml by LC-MS / MS method in the serum samples taken from the volunteers. In the present study, when chemiluminescence and HPLC methods are compared, ICC value was 0.911 (p=0.001), it was 0.924 (p=0.001) for chemiluminescence and LC-MS / MS methods, and was 0.961 (p=0.001) for HPLC and LC-MS/MS methods. The highest correlation was found among HPLC and LC-MS / MS methods, as well as it was observed that in all three methods is harmonious. Our study suggested that the measurement of vitamin D performed by all three methods is reliable. Therefore, HPLC, chemiluminescence or LC-MS / MS methods can be used safely to measure the levels of vitamin D in routine laboratories.
