Investigation of the effect of dexmedotymidine on TRPM2, TRPA1 channels and oxidant, antioxidant system after ischemia reperfusion injury in rat kidney

Abstract (EN)
Renal ischemia and reperfusion injury is a potentially serious problem seen during various medical and surgical interventions such as thrombolytic therapy after thrombosis of the renal arteries, organ transplantation, total circulatory arrest and cardiopulmonary bypass. The basic pathophysiology of I/R injury is an oxidative process due to microvascular dysfunction after reperfusion of ischemic tissues. This condition is clinically important because it is common and has a high mortality rate. In this study, it was aimed to examine the effects of using dexmedotimidine (DEX), TRPM2, TRPA1 channels on the expression activated oxidative stress by oxidant and antioxidant system and the effects on the apoptotic index calculated using the TUNEL method in rats with a kidney I/R model. In the study, 35 8-10 week old male Wistar albino rats were used. Experimental animals were divided into 5 groups with 7 animals in each group. No application was made to the control group during the experiment. In the sham group, the abdomen was opened under general anesthesia, the right kidney was removed and the left kidney pedicle was exposed; however, renal clamp was not applied. Dexmedetomidine-receiving group (DEX) was administered dexmedetomidine (100 μg/kg) at the onset of general anesthesia with the same surgical procedure as in Group 2. In the I/R groups, the rats were subjected to the surgical procedures described above and left kidney ischemia for 30 minutes followed by reperfusion for 45 minutes by applying a renal clamp to the left kidney peduncle. Dexmedetomidine was not given to the I/R4 group, but dexmedetomidine hydrochloride was given to the I/R5 group at the beginning of reperfusion. At the end of the experiment, the rats were decapitated by taking serum samples and kidney tissues under anesthesia. TAS and TOS values, TRPM2, TRPA1 expression were studied from both serum and kidney tissue taken from rats. TRPM2, TRPA1 expression and TUNEL method were studied by immunohistochemical method from the taken kidney tissue. While a statistically significant increase was observed in TRPM2, TRPA1 expression and TOS values in all serum and tissues taken in the I/R administered group (I/R4) compared to the non-administered groups, a decrease was observed in the TAS value. While a statistically significant decrease was observed in TRPM2, TRPA1 expression and TOS values in all serum and tissues taken in the I/R5 group compared to the I/R4 group, an increase was observed in the TAS value. There was no statistically significant difference in all values studied between the I/R5 group and the groups that did not undergo ischemia reperfusion.
