Abstract (EN)

ABSTRACTThe purpose of this study was to characterize the time course of renalischemia-reperfusion injury and the role of renal nerves on renal functions in the ratkidney. We also examined the effect of renal denervation and different time courseof renal ischemia-reperfusion injury on serum nitric oxide levels.Thirty-six male wistar rats were randomized into six groups. All ratsunderwent right nephrectomy to create a single kidney model. Renal denervated andinnervated rats were subjected to renal clamping for 30 min and 60 min. Group I(n=6) only underwent right nephrectomy, group II (n=6) underwent rightnephrectomy and left renal ischemia by occlusion of the renal artery for 30 minutes,group III (n=6) underwent right nephrectomy and left renal ischemia by occlusion ofthe renal artery for 60 minutes, group IV (n=6) only underwent right nephrectomyand left denervation, group V (n=6) underwent right nephrectomy, left denervationand left renal ischemia by occlusion of the renal artery for 30 minutes, group VI(n=6) underwent right nephrectomy, left denervation and left renal ischemia byocclusion of the renal artery for 60 minutes. After reestablishment of blood flow 24-hurine collected from each groups and blood samples were taken for analysis of bloodurea nitrogen (BUN), creatinine, sodium, chloride, potassium and nitric oxide.Thefunction of kidney was assessed by glomerular filtration rate (GFR), and fractionalexcretion of sodium (FENa), following reperfusion (24hr).The serum creatinine and BUN levels were significantly higher in G-IIBUN(52±25 mg/dl)-Creatinin(0,8±0,3 mg/dl), G-III BUN(77±30 mg/dl)-Creatinin(1,3±0,6 mg/dl), G-V BUN(78±31 mg/dl)-Creatinin(1,2±0,6 mg/dl), and G-VI BUN(110±29 mg/dl)-Creatinin(1,8±1 mg/dl), when compared with the G-I. BUN(26±3 mg/dl)-Creatinin(0,52±0,06 mg/dl). There was a significant decrease in GFRin G-III (0,5±0,4 ml/dk), G-V(0,4±0,3 ml/dk), and G-VI (0,19±0,14 ml/dk) withoutsignificant change in G-II (0,8±0,3 ml/dk) when compared with the G-I(1,4±0,5ml/dk). FeNa was over 1 in groupV (%1,09±1,01) and group G-VI (%1,4±0,9). Theserum NO levels were significantly higher in G- V (520±129 mmol/L) and G-VI(539±129 mmol/L) when compared with the G-I (336±111 mmol/L).Our results indicate that at physiological conditions denervation doesn?t affectrenal functions but it is worsening the injurious effect of ischemia-reperfusion injury.Our data suggest that at ischemia reperfused kidney serum Nitric oxide level doesn?timmediately return basal level.Key words: ischemia reperfusion injury, nitric oxide, denervation
