Endotoxaemia, or sepsis, is a heterogeneous class of syndromes caused by a systemic inflammatory response to infection. This condition may lead to severe shock and multiple organ failure and is one of the leading causes of death in critically ill patients. Acute lung injury (ALI) or acute respiratory distress syndrome(ARDS) is a serious clinical problem with high mortality and frequency. At last advertisements,it was shown that has been the role of NO, reactive oxygen species and cytokine on the pathogenesis of endotoxin-induced acute lung injury.Attempts to attenuate lung injury have focused on modulation of the signaling pathways leading to increased inflammatory cytokine/chemokine production, and on restoration of the oxidant/antioxidant balance to limit the degree of oxidative cell damage.İn the present study, intraperitoneally standart E.Coli injected rats were examined for the formation mechanisms of ALİ and the effects of intratracheal administration of NAC, methylprednisolone and vitamine complex on ALİ. We formed four experimental rat groups on this study. After intraperitoneal injection of E.Coli, ALİ has formed. At the end of the four hour, intratracheal NAC, methylprednisolone and vitamine complex have been applied.Tissue morphology andthe distribution of TNF-?, IL-6 and IL-10 have been examined in the lung sections.At the and of the study,it?s seen that vtamine complex and methylprednisolone reduced IL-6. Thus, both applications wereconsidered to be when used with good prognose in the early stage of ALİ.When vitamine complex group compared with NAC group and control group, it reduced IL-6 levels significantly (P<0.01).Methylprednisolone reduced IL-6 levels when compared with NAC group and control group. Although there has not been any istatically important difference on IL-10 levels,the application of NAC methylprednisolone seens to be more significant for providing protection. Vitamine complex group reduced TNF-? levels significantly when compared wih other groups (P<0.001). As methylprednisolone reduces IL-6 and increases IL-10 levels, when compared with NAC group and control group,it is considered that methylprednisolone can be preferred in the intratracheal administrations.Keywords: Acute lung injury, sepsis, NAC, metylprednisolon, vitamine complex, TNF-?
