Glutamate reuptake parameter changes and new treatment recommendations in experimental alzheimer model formed rats

Abstract (EN)
Aim: We aimed to investigate the measurement of glutamate reuptake times in rats with experimental Alzheimer's disease model by voltammetry, to correct differences with new treatments and to study the effects of these manipulations on clinical recovery. Material and method: Study plan was formed as 4 groups each group containing 6 rats. Intracerebroventricular streptozocin injection was performed in rats to establish an experimental Alzheimer's Disease model. Groups were determined as control group, streptozocin group, streptozocin + memantin group, streptozocin + ceftriaxone group. Each animal was subjected to behavioral tests, 2 days of passive avoidance and 5 days of Morris Water Maze tests. Glutamate reuptake times were measured by voltammetry. Statistical analyzes were performed using the One-Way ANOVA test through the SPSS 20.0 program. Results: According to our behavioral tests, on the 2nd day of passive avoidance test results were evaluated and it was found that learning and memory were disrupted in streptozosin group and this disruption was corrected in treatment groups. Similarly, in Morris's water tank test, the streptozosin group was shown to be defective compared to the control group. It has been shown that this disorder is corrected in treatment groups. According to the voltammetric test results, glutamate reuptake times in the prefrontal cortex and hippocampus brain regions were significantly longer in the streptozocin group and the prolonged reuptake times were significantly corrected in treatment groups. Conclusion: For the first time we have shown that there is a correlation between glutamate and Alzheimer's in this study by second by second measurements, and we have shown for the first time that memantine which is used in Alzheimer's treatment improves glutamate reuptake parameters. We have also shown that the ceftriaxone which is known to accelerate glutamate carriers is useful in the streptozocin-induced Alzheimer's disease model in animals.
