Investigation of effects of agomelatine in experimental renal ischemia/reperfusion injury

Abstract (EN)
Aim: Acute Renal Failure is clinically common situation caused by accidents, iatrogenic traumas, aortic clamp. This study aimed to investigate if agomelatine, one of melatonine analogues, which partially explained about its antioxidant and antiinflammatory effects has any scavenging role in experimental renal ischemia/reperfusion injury. Material and Method: In this study 60 Sprague Dawley type male rats used (n=60). Control group didn't have any application. 45 minutes ischemia and 24 hours reperfusion applied on I/R group. Same surgical procedure applied on Ago20 (K), Ago40 (K), Ago20 (U), Ago40 (U) groups. I/R group orally gavaged with hydroxiethylcellulose, other groups orally gavaged with 20 and 40 mg/kg concentrated agomelatine before ischemia. At the end of the study SOD, CAT, GSH, GPx and MDA parameters analysed on renal tissue and urea, creatin, IL-1β and TNF-α parameters analysed with serum tissue. Results: Depending on I/R injury increase of urea, creatine, IL-1β and TNF-α in serum tissue decreased by agomelatine treatment (p<0.05). On the other hand a decrease of SOD, CAT, GPx and GSH levels of renal tissue depending on I/R injury, increased by agomelatine treatment (p<0.05). In addition decreased MDA levels of renal tissue showed an increase with agomelatine treatment too (p<0.05). Conclusion: These results demonstrate that agomelatine is protective as an antioxidant and has scavenging free radicals effects against renal damage induced by ischemia/reperfusion.
