Investigation of The Effects of Saxagliptin on Experimental Renal İschemia Reperfusion İnjury Aim:Saxasagliptin is a dipeptidly peptidase-4 (DPP-4) enzyme inhibitör, pharmacological drug that has recently begun to be used in the treatment of diabets. Recent studies heve investigated the effects of saxsagliptine other than its protective role in diabets.It has been reported that saxsagliptin has antioxidant effects in experimental tissue damage in different tissues. In this thesis,the aim of study was to explain the role of saxsagliptine in experimentally induced renal İ/R injury in rats. Material and Method: Forty male Spraque-Dawley rats were used in the study.The rats were divided into for groups as Group 1 (control), group 2(ischemia-reperfusion; I/R), group 3 (İ/R+saxsagliptin 2mg/kg) and, group 4 (İ/R+saxsagliptin 10 mg/kg) were divided into 4 groups (n=10). No application was made to the control group.I/R group of kidney tissues bilaterally for 45 minutes ischemia and reperfusion for 24 hours. 1 hour prior to ischemia orally saline (saxsagliptine solvent) given orally. The rats in groups 3 and 4 received all I/R surgical procedures but 1 hour before ischemia in group 3 rats 2mg/kg in group 4 rats 10mg/kg saxagliptin was administered orally. When the reperfüzyon was over all ratswere sacrificed and kidney and blood tissues were collected. Conclusion: BUN and creatine levels in serum İ/R group increased compared to control group increased and it was observed that saxsagliptin administration significantly reduced (p<0.05). On the other hand, due to ischemia, MDA, KIM-1, NGAL and IL-18 levels increased in renal tissue due to ischemia (p<0.05), it was determined that MDA, KIM-1, NGAL and IL-18 levels decreased with saxsagliptin administration (p<0.05), again due to ischemia with decreasing GSH levels and SOD, CAT, GPx, enzym activity it was determined that the increase of saxsagliptin (p<0.05). As a result of histological analysis revealed that saxsagliptine reduced renal tissue damage (p<0.05). Results: The experimental study of saxagliptin,is a protective effect against ischemia-reperfusion injury it has been determined.
