The effects of vitamin D on adropine and betatrofine in experimental diabetic rat kidney

Abstract (EN)
Diabetes Mellitus (DM) is a heterogeneous disorder of metabolism characterized by hyperglycaemia. DM; is a chronic metabolic disease with microvascular and macrovascular complications, with accelerated atherosclerosis and along disorders of carbohydrate, lipid and protein metabolism. In this study; the effects of vitamin D on adropine, betatrophine and apoptosis in rat kidney tissue were investigated in streptozotocin (STZ) induced experimental diabetes model. 42 male Wistar albino rats aged 8-10 weeks were divided into 5 groups as 7 animals in each group. The control group had no application. A single dose of 0.1 M sodium citrate buffer was applied to the citrate buffer group. Vitamin D was administered 200IU / day orally to Vitamin D group. For the diabetes group, 50 mg / kg single dose STZ was injected ip which was dissolved in 0.1 M sodium citrate buffer. Rats with a glucose level above 250 mg/dl were considered diabetic. To Diabetes+Vitamin D group, 50 mg / kg single dose STZ which was dissolved in 0.1 M sodium citrate buffer was injected ip. Vitamin D was administered orally 200 IU / day after the formation of diabetes. At the end of the experiment, the rats in all groups were decapitated and kidney tissues were rapidly removed and TUNEL stained with immunohistochemistry. Total antioxidant status (TAS) and total oxidant status (TOS) levels were also investigated in serum samples from all groups. In biochemical and histological examinations, Serum TOS, TAS level and TUNEL positivity, adropine and betatrophine immunoreactivity were similar in the control, buffer and vitamin D groups. When compared with the control group, TOS level and TUNEL positivity increased significantly in diabetic group, whereas TAS level, adropin and betatrophine immunoreactivity decreased significantly. Compared with diabetic group, in Diabet+Vitamin D group TAS level, adropin and betatrophine immunoreactivity were significantly increased while TOS level and TUNEL positivity decreased significantly. As a result, it has been concluded that further studies should be performed to try vitamin D-related treatment approaches to prevent complications of diabetes and to investigate the adropine and betatrophine in the pathogenesis of diabetic nephropathy.
