The effects of Ankaferd on the experimental model of hemorrhagic cystitis

Aim: We investigated the effect of Ankaferd on cyclophosphamide (CP) induced hemorrhagic cystitis (HC) in rats.Materials and Methods: Twenty four female Wistar rat ages between 8-12 weeks were divided into three groups by random sampling method. Group 1 (Control) animals were injected with the same amount of saline. Group 2 (CP) received only 100 mg/kg urotoxic dose of CP, Group 3 (CP+Ankaferd) received CP and Ankaferd. Ankaferd was given in dosage of 0,5 cc/kg intravesical one hour before CP injection and 12, 24, 36, 48, 60 hours after CP injection. The bladder tissues were cut into two equal pieces from top to the bottom. One half was stored at -80 0C for determination of SOD and MPO activities with MDA and NO concentration and the other half was fixed for 24 hours in 10 % buffered formaldehyde for histological evaluation.Results: Cyclophosphamide injection which caused HC increased MDA levels indicating oxidative stress, while Ankaferd reduced MDA levels in the bladder and this was statistically meaningful. SOD, an antioxidant enzyme, activities increased in the tissue samples of CP+Ankaferd group as a meaningful. MPO activities decreased more significantly at the group treated by Ankaferd than CP group. There was no difference between three groups at the point of NO levels. Control animals had histologically normal bladders except edema. The reason of that edema was estimated as the trauma happened during expulsion of the bladder. CP caused severe microscopic bladder mucosal injury. Ankaferd didn?t show histological protection against bladder damage of CP. The ratio of the bladder weight to total body weight increased at the CP+Ankaferd group more significantly than CP group. This result is convenient to bladder histology. An important difference about hematuria was not found between CP and CP+Ankaferd groups.Conclusion: New studies about the dosage and usage time of Ankaferd are needed to have better clinical and histopathological results.Keywords: Hemorrhagic cystitis, cyclophosphamide, Ankaferd, rat, oxidative stress.
