Abstract (EN)
In this study, against to DMBA-induced breast cancer healing effects of Pistacia terebinthus L. subsp. palaestina and Rhus coriaria L. were examined as biochemical, molecular and histopathological, in the female Sprague-Dawley rats. 66 rats were used in this study. Rats were divided into 6 groups as Control, DMBA, MN, SM, MN+DMBA and SM+DMBA. DMBA was administered by gavaged, a single dose of 80 mg/kg according to body weight, to 8-week-old rats. Aqueous extracts of terebinth and sumac were given orally 3 days a week to rats in antioxidant groups. In biochemical studies, total protein, MDA, GSH, fatty acid and vitamin levels and changes in activities of CAT, GST and SOD which are antioxidant enzymes in breast, brain, liver and kidney tissues were examined. Total protein levels generally decreased in DMBA group compared to control (p<0.05).While the levels of MDA in DMBA groups of all tissues increased compared to the control group; decreased in antioxidant groups (p<0.05). Lipophilic vitamins and cholesterol levels were analyzed by HPLC. The levels were different in DMBA and antioxidant groups of tissues. Fatty acids were analyzed by GC. As a result of analysis, acids such as palmitic, palmitoleic, stearic, oleic, linoleic, arachidonic, docosahexaenoic were found to be high in tissues. These fatty acid levels showed different changes in DMBA and antioxidant groups of tissues. In molecular studies, expression levels of HER2, P53, Bcl-2, Bax, Caspase-9 and Ras proteins were analyzed by western blot technique. In histopathological studies, breast tissue sections were prepared in hematoxylin-eosin dye solution and examined under microscope. The sections were examined in terms of features, such as tumor type, diameter, differentiation, fibrosis, necrosis. As a result, oral consumption of sumac and menengic aqueous extracts was found to be effective in rats with breast cancer.
