Determination of therapeutic effects of tamoxifen and sodium thiosulfate in rats with experimental xanthium strumarium poisoning

Abstract (EN)
Attractyloside (ATR) in pıtrak (Xanthium strumarium) is toxic especially in liver and kidney tissue and causes cell death as a result of oxidative stress and mitochondrial dysfunction. Tamoxifen (TAM), an agent used against some types of cancer in living beings, also prevents mitochondrial pore opening and formation of reactive oxygen species in cells. Sodium thiosulfate (STS), on the other hand, is a liver-protective compound as a source of sulfur as well as preventing the production of ROS. In this study, the therapeutic effects of TAM and STS on these tissues were investigated in rats with liver and kidney damage with pıtrak. 155 female Sprague-Dawley rats were used in the study Rats were divided into 7 groups (n=7). TAM and STS were given to the rats at the 6th hour after the pıtrak was given, and the application was repeated at the same time every day. At 6, 24, 72, 120 and 168 hours following the administration of the treatment agents, 5 animals from each group were decapitated and their blood, liver and kidney tissues were collected. Compared to the control group, the levels of AST, ALT, ALP, LDH, CK, BUN, creatinine and MDA increased in the pıtrak group; It was determined that glucose levels and GSH and SOD activities decreased. In addition, mitochondrial Ca+2 level, mPTP opening rate and ATP-synthase activity increased; histopathological examinations revealed an increased necrotic cell death rate. It was determined that the treatment groups brought oxidative stress and blood biochemistry parameters closer to normal. While ATR+TAM and ATR+TAM+STS groups had a positive effect on mitochondrial Ca+2 increase and mPTP opening, it was observed that ATR+STS did not have any effect on mPTP. While a positive effect was observed in all three treatment groups in preventing pathological damage, it was determined that this effect did not occur in all time periods. Increased ATP-synthase activity was decreased in all three treatment groups. The findings have shown that TAM and STS compounds can be helpful in the treatment of pıtrak poisoning, especially when used together.
