Effects of deferoxamine on acute kidney injury in experimental sepsis model

Abstract (EN)
Sepsis is an increasingly important cause of mortality and morbidity all over the world, with an annual incidence of 2.4 – 3.0/1000 cases. Clinically, in the sepsis patient group; one of the most important factors affecting mortality and morbidity is acute renal failure. Deferoxamine is an iron chelator and free radical scavenger. The aim of this study is to investigate the possible protective effects of deferoxamine (DFX), which is known to have antioxidant effects, on sepsis-related acute kidney injury. Application of lipopolysaccharide (LPS) is a frequently used and safe method to create an experimental sepsis model. In this context, 30 Wistar Albino type rats were divided into 5 groups: control group, LPS group, DFX+LPS group, LPS+DFX group, DFX group. LPS was administered to the LPS group at a dose of 15 mg/kg and kidney tissue and blood samples were taken 24 hours later. In the DFX+LPS group, 20mg/kg DFX was administered every 24 hours and the second dose of DFX and LPS was administered. Tissue samples were taken after 24 hours. DFX application was applied to the LPS+DFX group at the same time as LPS application. The second dose of DFX was administered 6 hours later and tissue samples were taken 18 hours later. When examined in biochemical results; Statistically significant changes were detected in IL-6, MDA, NGAL, SOD, TNF alpha, IL-1 beta and GPx measurements with DFX application. In histopathological examination, protective effects of DFX application on Bowman capsule dilatation and tubular epithelial degeneration were observed. In conclusion; it has been concluded that deferoxamine has protective effects against acute kidney injury due to sepsis.
