Diagnosis and statistical evaluation of klebsiella pneumoniae using molecular methods from some hospitals in Dhi-Qar governorate (Iraq)

Abstract (EN)
In this study 386 samples were collected from hospitals in Dhi-Qar (Iraq)with ages 10-60 years old between July and September 2022. The samples were identified; 69 withKlebsiella spp. The others were Escheirechia coli, Staphylococus aureus,Pseudomonas spp. and Proteus, S. epidermidis, and Enterobacter spp. The API 20E and VITEK 2 system were used to identify the biochemical tests. The disc-diffusion method assessed the antibiotic sensitivity of Kleibsila pneumoniesamples against 15 rourenary tract infeictionnely used antibiotics. According to the results, the observed spectrum of DNA concentrations ranged from 47.4 to 123.8 ng/L, as well as the integrity of the genetic material (1.6-2.0).Polymerase chain reaction was used to study distribution pathway infection of 16S r-RNA, rmp-A, serogroups to it (kfu genes) and molecular detection of Kleibsila pneumonia samples.29 (60.4%) of the samples are of the K-1serotyp, yielding a band of 1283 bp in size, and 14 (29.2%) are of the K-2serotyp, yielding a band of 543 bp in size. The findings of this study indicate that the magA and k-2A genotype may be a good biomarker for identifying K-1 and K-2serogroups of Kleibsila pneumonie, as these serogroups were significantly extraabundant than the non-K-1/K-2serotyps (Non-K-1/K-2; 5 swabs, 10.4%). The present study employed a dataset comprising 366 clinical swabs. The dataset encompassed diverse parameters, including patient's identifiers, age, gender, pre-existing medical conditions, antimicrobial resistance, residential locality, educational attainment, and smoking patterns. A statistical examination was conducted on the collected data for each of the aforementioned variables to evaluate the predisposition of patients to K. pneumoniae infection.Whether there is a statistically significant relationship between the variables considered was tested with the Chi-Square test. According to the Chi-Square test, a statistically significant relationship was found at 0.05 significance level between smoking-antibiotics resistance, housing-antibiotics resistance, and education-antibiotics resistance variables.
