Studies on synthesis and in vitro cyclooxygenase inhibitory activities of diaryl heterocyclic compounds

Abstract (EN)
In this study, twenty-eight original of thirty-two compounds having 2-oxo-5H-furan, 2-oxo-3H-1,3-oxazole and 1H-pyrazole as central ring while 3-methyl-2-oxo-3H-benzoxazole and 4-sübstitutedphenyl attached were synthesized. Compounds were differentiated by substitution of phenyl ring with small groups such as OCH3, Cl, F, SO2CH3 at para- position.Based on the literature, the compounds synthesized are expected to have COX inhibitory activity. The activities of the final fifteen compounds for possible COX inhibitory activity were performed by in vitro Purified Enzyme Assay at 10 µM concentration and the IC50 values of the active compounds were calculated. A 2-oxo-5H-furan derivative Compound 7 and 1H-pyrazole derivatives Compound 25, 26, 27 have COX-2 IC50 values as 0.325, 0.011, 0.011, 1.000 µM; COX-1 IC50 values as 0.061, 0.004, 1.000 and 6.000 µM, respectivelyMolecular modelling studies which were performed on the binding site of COX-1 and COX-2 using the compounds having COX inhibitory activity, support the results of in vitro enzyme assay.
