Evaluation of TNF-α fractalkine and TGF-β chemokin levels effective on gingival overgrowth

Abstract (EN)

Our aim in this thesis project is to evaluate the presence of fractalkine levels in DOS and gingival tissues of patients with gingival overgrowth plaque -induced and drug -induced (amlodipine), to determine TNF-α, fractalkine and TGF-β levels, and to examine the relationship between these findings and clinical periodontal parameters. This study is the first study investigating the fractalk in gingival overgrowth and gingival tissue. A total of 45 individuals with plaque-induced gingival overgrowth (P) 17, drug (Amlodipine) induced gingival overgrowth (I) 18, control group (K) systemically healthy, without gingival overgrowth, 10 with aesthetic crown lengthening the individual was included in the study. Plaque index (PI), gingival index, (GI), pocket depth (CD), probe bleeding index (SC), clinical attachment level (CAS), gingival overgrowth index (DBI) were measured to determine the periodontal health status. DOS and tissue samples were collected from all individuals included in the study. ELISA was used to determine the concentration of TNF-α, fractalkine, and TGF-β in DOS samples, and Real-Time qPCR method was used in tissue samples. As a result of the statistical analysis, tissue fractalkine and TNF-α levels were found to be significantly higher in the P group compared to the I group (p <0.05). TNF-α level in group I was found to be significantly lower than group K in DOS. The mean PI, GI, CD, SK, DBI values of all mouth P and I groups were found to be significantly higher than the control group (p˃0.05). It was found that the mean PI in the sample area was significantly higher in the P group compared to the I and K groups. Sampling area; The mean GI, CD, SK and DBI values of the P and I groups were found to be significantly higher than the K group (p˃0.05). Strong positive correlation between TNF-α and TGF-β levels (r = 0.770), TNF-α and Fractalkine levels (r = 0.745), and TGF-β and Fractalkin levels (r = 0.647) in P group in DOS is observed. A strong positive correlation is observed between tissue TNF-α and TGF-β levels (r = 0.697), between TNF-α and Fractalkine levels (r = 0.689), and between TGF-β and fractalkine levels (r = 0.903) (p <0 , 05). In group I; There is a strong positive correlation between tissue TNF-α - TGF-β (r = 0.773), TGF-β and fractalkine levels (r = 0.762) (p <0.05). In our study, the increased fractalkine and TNF-α levels observed in tissue samples in the P group compared to the K group suggest that fractalkine may play a role in gingival overgrowth and inflammation in periodontal diseases.
