Diesel exhaust particles, which exits by burning diesel fuel, consist of a considerable amount of particular matters wandering in the urban. There is a quite close relationship between diesel exhaust particles and very crucial health problems such as chronic obstructive pulmonary disease, brutal asthama, being sensitive against respiration infection and pulmonary inflamations. On the other hand, the contrivance of pulmonary diseases thanks to diesel exhaust particles has not ever been explained clearly. In this work, cell cycle of A549 cells of which human primer bronch epithelial cells of diesel exhaust particles and lung alveolar cell arrays and also the affect of genes that order apoptosis to mRNA expression levels have been researched. Primer bronch epithelial cells acquired from smoking and non-smoking patients with chronic obstructive pulmonary disease were exposed to three different study groups who are free-serum culture medium, 10 ? g/ml DEP ve 100 ? g/ml DEP. Pulmonary alveolar cell sequences that are A549 cells were exposed to free-serum culture medium, culture medium with serum (10% FCS) and 50 ? g/ml DEP. Besides, study groups which include JNK and ERK inhibitories (10 ?M, 33 ?M) and n-acetylcysteine (NAC33 mM) in the substances of 50 ? g/ml of DEP were added up and examined how these elements effect the affect of DEP. cDNA samples obtained from these cells with cell cycle and silkinE that regulates apoptosis, siklin dependent kinase inhibitory P21, transcription factor NF- ?b, proto onkogenic c-MYC, MAPK path regulaters Jnk2 ve Erk2 genes mRNA expression levels were studied by the method of RT-PCR. Moreover, after the DEP exposition of the genes, the release of interleukin (IL)-8 and granulocyte-macrophage colony-stimulating factor (GM-CSF) was researched by the method of linked immunosorbent assay (ELISA). In the conclusion of this study, what was learnt is DEP might make a remarkable change in the expression levels of mRNA, that human primery bronchial epithelial cells and pulmonary alveolar cell sequences that are A549 cells cell cycle apoptosis regulator.
