Abstract (EN)
Due to the high incidence of triple-negative breast cancer in women globally, which is characterized by poor diagnosis and treatment, there was a need to search for new and innovative ways to diagnose and investigate this type of breast cancer at the epigenetic level. Epigenetic inheritance is the key to understanding diseases and the physiological changes in the organs and tissues of the body, and global and normal precancerous breast cancer in particular. DNA methylation plays an essential role in the pathogenesis of diseases in the human body. Understanding gene expression, epigenetics, and DNA methylation and their relationship to the ING1 tumor suppressor gene family can contribute to the development of treatment for triple- negative breast cancer. Through our research, we investigated the relationship between methylation of the ING 1 promoter site and triple-negative breast cancer. As a result of this study, we found the relative methylation rate was 93.5% in our triple- negative breast cancer samples. We noticed an increase in DNA methylation in triple breast cancer cells, which may be related to its expression decreasing in this aggressive breast cancer subtype. We suggested the ING-1 gene may be a potential marker for the early diagnosis of triple-negative breast cancer. We used 92 samples taken from women diagnosed with triple breast cancer with different stages of cancer. We found a close relationship between DNA methylation and this type of breast cancer. We also found a decrease in the expression of the ING-1 gene in the cells. As 93.5% of the samples showed the presence of DNA methylation, and only 6% showed un-methylation.
