Abstract (EN)

In this study,submerged fermentation and optimization of gluconic acid was performed by using Aspergillus spp. strains isolated from moldy food materials such as onion peel, garlic peel,bread and dextrose monohydrate as substrate.The qualitative and quantitative analyzes of the fermentation product were performed on HPLC to determine the optimum conditions for gluconic acid fermentation. By using natural isolatedAspergillus sp.optimum conditions for the production of gluconic acid was determinated as at 600 rpm, pH 5.5, 30oC with addition 0.5 vvm pure oxygen on the 7th day including in fermentation medium containing10% dextrose monohydrate. As a result of the optimization, gluconic acid production was obtained with a yield of 69%.In addition, it was observed that the addition of 3000 units of glucose oxidase and 500 units of catalase to the fermentation medium reduced the fermentation time to 3 days and yielded 95% yield of gluconic acid. According to these results, it can be said that using by natural isolated Aspergillus sp. almost completely of dextrose monohydrate converted gluconic acid under the determined optimum conditions.Gluconic acid is a product that can be used safely in many sectors such as construction, agriculture, health, food and can be easily biodegraded in nature.
