Abstract (EN)
Hematopoietic stem cell transplantation (HSCT) is a standard treatment for bone marrow reconstruction after leukemia cells are destroyed by drug therapy. Natural killer cells after HSCT are the first group of cells to be engraftment in the bone marrow. NK cells are clinically important because they can reduce the occurrence of post-transplant infection and reduce the formation of GVHD, in addition, anti-leukemic effects have been described in different hematological diseases. Different studies have been designed for the expansion of NK cells in vitro applications. There is a need to create alternative nutritional and support medium in cellular therapy applications. For this purpose platelet lysates are used effectively in cell proliferation. Through the formation of lysate the factors in the platelet granule are released into the medium. Our aim is to evaluation of the effect of cord blood platelet lysates on natural killer cells. Cord blood was collected from volunteer pregnant women then MNH isolation and natural killer cell selection were performed and platelet lysate was prepared. NK cells were stimulated in the presence of IL-2 and platelet lysate under cell culture conditions. Differences between groups were evaluated by flow cytometric method and automatic cell count. To our knowledge, this is the first study to demonstrate the efficacy of cord blood derived platelet lysates on natural killer cells. NK cells are usually can proliferate after prolonged incubations in vitro. There is a need to use multiple cytokines during the NK stimulation phase. This study demonstrated for the first time that platelet lysates can provide NK proliferation in a short time with IL 2. However, incubation time should be extended and supported by studies in which changes in platelet lysate concentrations are tested.
