Abstract (EN)

In this study, highland and lowland regions of Adana, Mersin and Hatay provinces of the Eastern Mediterranean Region were surveyed to collect infected fruit with Ceratitis capitata Wied. (Diptera: Tephritidae). As a result, C. capitata individulas were obtained from 27 different orchards an thet were used in molecular systematics analysis. Mitochondrial DNA cytochrome oxidase (CO) gene were used for phylogenetic analysis. The isolated genomic DNA samples was amplified by using PCR, totaly 914 bp nucleotide sequences of COI-COII region were obtained via C1-J-2792 (George) and TK-N-3772 (EVA) primer pairs. 72 samples were sent for sequencing. Out of 67 samples were give good reading and after that alligment and correction of sequences by Mega6.0. Sequences were analyzed to obtain number of haplotypes by using DnaSP 5 which is calculated seven haplotypes. Arlequin program were used in calculation of gene flow (Nm) and binary genetic distance (Fst) values. Genetic differences between populations in lowland and highland parts of Adana, Mersin and Hatay provinces were assed and 0.00486 value obtained as highest Fst values between Hatay and Mersin Provinces, and lowest one observed as -0.01604 between Hatay and Adana. Nm value was observed as limitless (∞) between Adana and Mersin provinces but 604.84 was seen between Hatay and Mersin. These values clearly show us that; the population of Adana and Mersin provinces has mixed population of C. capitata compare to between Hatay and Mersin provinces. Haplotytpes of highland population of three provinces is become differeent each other. Moreover, genetical diversity (Hd) in populations of higher elevations is greater than the populations in lowland areas. Phylogenetic tree drawn by existing examples with referance gene of COI-COII region which are obtained from NCBI website and they are the populations of China, USA, Australia, Greece show that Turkey's poulation has different branch than other countries, and furhermore highland population has aslo different branch in Turkey populations.
