Abstract (EN)

THE EFFECTS OF VITAMIN D ON SPEXIN IMMUNOREACTIVITY IN KIDNEY TISSUES OF DOXORUBICIN-TREATED RATS Doxorubicin is an anthracycline derivative antibiotic with widespread use including chemoterapy, but its use in the clinic is limited due to its side effects. It is a neuropeptide belonging to the spexin galanin family and has effects on energy metabolism. Vitamin dis an antiapoptotic antioxidant and immunomodulatory agent that reduces oxidative stress. The aim of this study was to investigate the effects of vitamin D on spexin immunoreactivity in kidney tissues of doxorubicin treated rats. In this study, 28 male Spraque-Dawley rats aged 8-10 weeks were used. The experimental animals were randomly divided into 4 groups with 7 animals in each group. No treatment was applied to the control group during the 14-day study period. Rats in the vitamin D group received 200IU/day vitamin D orally. In the doxorubicin group, a single dose of 10 mg/kg doxorubicin was administered intraperitoneally (i.p.). Rats in the doxorubicin+ vitamin D group received a single dose of 10 mg/kg doxorubicin ip. vitamin D 200IU/day orally every day during the 14-day experimental period. At the end of the experiment, rats were decapitated and blood samples for biochemical analysis and kidney tissues for histological evaluations were taken under appropriate conditions. Total antioxidant status (TAS), total oxidant status (TOS) and spexin levels were determined by ELISA method in the sera of the experimental animals. In addition, in order to determine spexin immunoreactivity in kidney tissues by immunohistochemical method. As a result of the study, Spexin levels in both kidney tissues and serum samples of rats from control and Vitamin D groups were statistically similar. However, these levels were statistically increased in doxorubusin group compared to control group. On the other hand, in the doxorubusin+ Vitamin dgroup, which was the treatment group, spexin levels were statistically decreased compared to doxorubusin group. And also TAS/TOS parameters used in the determination of oxidative damage were similar in the control and Vitamin D groups, while serum TOS level increased and TAS level decreased statistically in the doxorubusin group compared to the control group. In doxorubusin+ Vitamin dgroup in which vitamin D was given as treatment, serum TAS level increased and TOS level decreased statistically. In conclusion, Doxorubicin administration increased TOS and spexin levels and decreased TAS levels, while vitamin Dgiven as treatment decreased TOS and spexin levels and increased TAS levels. It was observed that vitamin d suppressed oxidative stress against Doxorubicin -induced renal damage and regulated spexin levels. It was concluded that further and more comprehensive studies are needed to elucidate the role of spexin in oxidative damage caused by chemotherapeutic response.
