In parallel to developments in human beings' life their interest in the music and desire tomanage the sounds also developed throughout the course of time. Discovery of new instruments and their evolvement had boosted their interest to create new compositions in a way to their own wish through with managing the sounds. In this context, it was inevitable use of technological devices in creating new sound synthesis. Even though the VST synthesisers are being used in today's musical studies, it is a question in minds that how the musical instruments were replaced with them and similarities between them. For that reason, in this research it was aimed to examine the differences between instrumental and VST synthesisers. It is believed that the outputs of this research will help to the people who have been working in the field of musical technologies. In this research "Matlab" software was used to collect THD data from oscillator sounds produced by instruments and VST synthesisers. Also "Voxengo Span" software was used to get spectrum analysis findings from the both of them. Within this study a melody with three different sounds (simple-with effect and arranged) was prepared by using musical instrument and VST synthesiser separately. Then a semi-structured questionnaire was used to collect data from 20 sound engineer and 20 arrangers participated in the research about how they interpret the three different sounds of the melody (simple-with effect and arranged). The results of the study indicated that most of the sound engineers, tonmeisters and arrangers preferred the MS-20, i Tal-U VST'i and Roland JX-8P instruments respectively. In another words instruments were preferred mostly by the participants of the research. In the context of reasons for preference; the results revealed that they choose the sound they liked and looked for the instrument/analog sound sensation.
