Abstract (EN)

The purpose of our study is to research the polymorphism rates in the short sequential iterative sequences of the 16 loci on the Y- chromosome of masculine individuals, who are members of the population in Elazığ. It has been aimed with the detected polymorphism to constitute Y-STR DNA banks which belong to the Turkish population, and to examine the sensibility of 16 Y-STR loci which belong to the population of Elazığ in forensic cases. It?s also considered that it will also have a share in enlightening some familytree determinations and mass migrations.This study was carried out at the DNA Laboratory of the Department of Forensic Medicines at the Firat University Faculty of Medicines, on blood species of local 250 masculine individuals in Elazığ who weren?t relatives of eachother. GenoMik DNA isolations were derived from blood species. The AmpF/STR Y-Filer TM PCR Amplification Kit of the isolated DNA were used for ingreasing the gens areas of the desired loci. The electrophoresis of PCR items was carried out with the ABI 310 Sequence Device, and the results were analyzed with GeneMapper Software Version 3.2. The haplotypes and densities which emerged after the results were evaluated with Arlequin 3.0, and were determined respectively.There are few studies related with Y-STR loci in Turkey (DYS456, DYS389I, DYS390, DYS389II, DYS458, DYS19, DYS385a/b, DYS393, DYS391, DYS439, DYS635, DYS392, Y GATA H4, DYS437, DYS438, DYS448), and none in Elazığ. Y-STR are unique for each community and are also unique for every ethnic society in the same country. Thus, considering that Y-STR densities which were determined abroad, were also valid for Turkey or applying processes based on these densities is wrong and displays a deprivation of any scientific basis. In our study, no special allele was found in Elazığ, and the highest genetic variety locus was DYS385a/b (0,962), the lowest was DYS391 (0,473). 15-16 dublication was detected in locus DYS19. 250 varieties of haplotypes were derived from 250 individuals and a sensibility of 1.0000 resulted when all the loci were taken into attention. It?s determined that the 16 Y-STR loci is reliable for paternity tests, and forensic cases because the sensibility for the population of Elazığ is high.As a result, the haplotypes, allele and haplotype frequences of locus 16 Y-STR were derived, and it?s supposed that contribution will be caused to the comparison of these data and the previous population studies, and enlightenment to the phylogenetic relations.Keywords: Y chromosome, short tandem repeat, haplotype, Elazig
