ABSTRACT INVESTIGATION OF SALMONELLA IN FRESH WATER TURTLE (Mauremys caspica caspica, Gmelin, 1774) CAUGHT FROM REGION OF ELAZI? CITY SEWAGE DISCHARGE INTO KEBAN DAM LAKE Masters Thesis Mikail ÖZCAN Fırat University Graduate School of Natural and Applied Sciences Department of Aquaculture 2004, Page: 52 In this study, Salmonella was isolated and identified by culture method in 25 alive freshwater turtle (Mauremys caspica caspica) caught from Koçkale Region of Keban Dam Lake. Where Elazığ City sewage discharged Salmonella isolates was also confirmed by means of Polymerase Chain Reaction (PCR) technique. For this, freshwater turtle liver and intestine prepared from homogenized stock pepton water after pre-enrichment Rappaport Vassilliadis Pepton Broth smear made on Birilliant Green Phenol Red Lactose agar, Xylose Lysine Tergitol 4 agar, MacConcey and Salmonella-Shigella agar selective mediums. Biochemical analysis and DNA extraction steps were applied to isolated Salmonella suspected agent. DNA extracted from the isolate were amplificated at PCR by using a pair of primer produced from 16S rRNA gene of Salmonella. According to this result, 6 out of 25 turtle were found efective with Salmonella from Koçkale region of Keban Dam Lake Salmonella spp. could not be isolated from 10 water samples from study area. The fact that, Salmonella causes important diseases in humans such as septicemia gastroenteritis, thyphoid and paratyphoid should be taken into consideration, in the light of the data obtained in this study, alive fresh water turtle in Koçkale Region was put middle became threat for human health with other water living and fish for give out feces Salmonella.
