Abstract (EN)

The previous Bokhman classification was used to classify the endometrial carcinoma and was found ineffective with the clinicopathological correlation. Therefore in 2020, the WHO changed their classification to the molecular classification. Molecular classification has one big challenge, the POLE mutation status only can be determined by molecular methods. Many hospitals around the globe aren't able to perform molecular methods. In a previous study, AMF (Autocrine Motility Factor) and AMFR (Autocrine Motility Factor Receptor) proteins were found to be increased in POLE mutant endometrial carcinoma cases. Therefore we applied AMF and AMFR immunohistochemistry to endometrial carcinoma cases to see whether this approach can find POLE-mutated endometrial carcinoma cases. Furthermore, we counted tumor budding and scored it into four different tumor budding scores to understand the relationship between epithelial-mesenchymal transition and AMF, AMFR antibodies. We applied AMF and AMFR immunohistochemistry to 55 endometrial carcinoma cases molecularly classified by Koca Yılmaz et al.. Then we examined the H&E slides of the 55 cases and counted the tumor buds. Later we transformed tumor bud counts into tumor budding scores. AMF antibody was able to reach a score of 6 and more on 9 out of 11 POLE mutant cases. Also, the AMF antibody reached a sensitivity of %81,8 and a specificity of %61,4. AMFR antibody showed low expression in high peritumoral tumor budding scored cases (p=0,043). As a result of our findings, we believe that immunohistochemical AMF application can be used to triage high-potential POLE mutant cases in the practice.
