The effect of endotoxin on microRNA expression profile in N9 mouse microglial cell line

Özet (EN)

Microglia is a type of glial cell that are known as macrophages of the brain. They are immunophenotypically and functionally similar to monocytes and macrophages. In the central nervous system (CNS), microglia plays an important role in the inflammatory process. MicroRNAs (miRNAs) which are small, non-coding RNAs have a crucial role in gene regulation at the post-transcriptional level. In this study we aimed to search the role of miRNA on microglial activation.In the experiment, N9 cells were treated with 10-1000ng/ml doses of Lipopolysaccharide (LPS). In preliminary study, by using 100 ng/ml LPS, N9 mouse microglial cells were activated. By using microarray method, miRNA expression changes were determined. Among miRNAs which were determined by microarray, the gene expression of miR-191, miR-193 and miR-431 were examined by qPCR. Afterwards, target genes of those miRNAs were determined by bioinformatics analysis. The validation of target genes which were determined before was done by Luciferase assay.According to qPCR data, its shown that the expression of miR-191,miR-193 and miR-431 increased. The validation of target genes (IL-6, IL-10 ve IRAK1 ) which were determined by bioinformatic method, were done. Of these miRNAs, it has shown that miR-191 targets IL-6, miR-193 targets IL-10 and miR-431 targets IRAK1.
