Protective effect of alpha-linolenic acid on LPs i̇nduced orchitis in mices

Introduction and Objective: Alpha-linolenic acid (ALA) which is one of polyunsaturated fatty acids is very important for human life. ALA is a protective nutrient for the heart and cardiovascular system. However, ALA has very strong anti-inflammatory and anti-oxidant properties. Increases in the expression of inflammatory medaiators are accompanied orchitis induced by lipopolysaccharide (LPS). Therefore, twe aimed to determine whether ALA, which has anti-inflammatory properties, is protective effect against LPS-induced orchitis. Material and Method: Mices were divided into three groups as control, orchitis and orchitis + ALA applied. Orchitis were induced by intraperitoneal LPS application in mice and treated with ALA than cyclooxygenase-2, phospholipase A2, inducible nitric oxide synthase (iNOS) enzymes and nuclear factor kappa-B (NFκB) were analyzed by Enzyme-Linked Immuno Sorbent Assay (ELISA) method in their testes, IL-6 and TNFα in their serum. Differences in results between tissues were tested by one way analysis of variance (ANOVA) and corrected by Bonferroni multiple-comparison test. Results: LPS administration to the mices increased the expression of inflammatory mediators such as IL-6, TNFα and NFκB and COX-2, FLA2 and iNOS enzymes. It was determined that ALA treatment significantly reduced these mediators and enzymes levels. Conclusion: This study showed that the ALA can be beneficial for LPS-induced testicular inflammation and testicular damage. This study will help the researcher to identify important points of testicular inflammation caused by Gram negative bacteria. Keywords: Orchitis, LPS, inflammation, alpha-linolenic acid, mice
