Comparative genome analysis of the multidrug resistant Mycobacterium tuberculosis isolates from different geographic regions

Abstract (EN)

The aim of this study is to find out genomic alterations playing role in drug resistance and possible differences between isolates belonging to different geographic regions. For this, comparative genome analyses of multidrug resistant (MDR) Mycobacterium tuberculosis (MTB) isolates, causing the disease tuberculosis, from different geographical regions were performed using non-MDR H37Rv strain (Genbank accession number AL123456.3) as reference. The genome analyses of the strain 410 from Argentina (Genbank accession number CM007646) and the strain PR10 from Malaysia (Genbank accession number CP010968) revealed 435 and 430 proteins with putative mutations, respectively. Majority of the mutant proteins were included into lipid, carbohydrate, protein, nucleic acid, and energy metabolisms, oxidation-reduction process, transport and membrane structures as well as proteins with unknown function. The number of common proteins in both MDR strains was 90; 11 in lipid metabolism, 6 in carbohydrate metabolism, 8 in protein metabolism, 16 in nucleic acid metabolism, 6 in redox process, 3 in energy metabolism, 3 in iron and sulfur metabolism, 10 in transport function, 1 in drug and xenobiotic metabolism, 8 in virulence, 2 in cofactor metabolism, 4 in signal transduction, 3 in membrane and 1 in cell wall structures, and 3 in diverse pathways. Consequently, it was determined that the putative mutant proteins were mostly related with biofilm production, DNA repair, replication, transcription and translation mechanisms, aerobic respiration, type VII secretion system, two-component signal transduction, polyketide synthesis, vitamin B biosynthesis, cell wall, membrane and transport functions.
