The in vitro Evaluation of Antimicrobial Properties and Genotoxic Potential of Different Dentin Bonding Agents

Abstract (EN)

The interest to bonding agents has been increased recently because of probable biological effects caused by the close and continual contact of dentin bonding agents with tooth and surrounding tissues. The aim of the present study was to evaluate the different dentin bonding agents by assessing antimicrobial property and mutagenic potential. In the first experiment, performed in this study, the antibacterial activity of Clearfil SE Bond, SL Bond, i-Bond, Clearfil Protect Bond, %2 chlorhexidine (positive control) and SF (negative control) against L.casei (ATCC4646), C.albicans (ATCC10231) and S.mutans (NCTC10449) was determined by agar disc diffusion test and serial microdilution assay. In the second experiment, the genotoxic activity of dentin bonding agents, was investigated at different elution concentrations in human lymphocytes using the alkaline single-cell gel electrophoresis technique. According to the results of agar disc diffusion experiment and serial microdilution assay, Clearfil Protect Bond primer, showed clear inhibition against all three bacteria among the other adhesive systems. The other systems tested in this study, Clearfil SE Bond produced a little antimicrobial effect on S.mutans and SL Bond showed inhibition against the three microorganism. i-Bond, produced significantly less antimicrobial effect against three microorganism. According to the results of genotoxicity assay, Clearfil Protect Bond primer, at 50 ?l and 100 ?l concentration increased the tail length significantly. Moreover, tail moment and tail intensity were significantly increased at 100 ?l concentration A significant increase in tail moment, intensity and length was observed at 100 ?l concentration for Clearfil Protect Bond adhesive system. Clearfil SE Bond primer at 50 ?l and Clearfil SE Bond adhesive at 100 ?l resulted in the increase of DNA damage. However SL Bond and i- Bond did not produce significantly DNA damage. The results of two experiments designed for this study, indicated that, among the adhesives tested, Clearfil Protect Bond, that contains antimicrobial monomer (MDPB), at clinically effective dose, have strong bactericidal/fungucidal effects and low genotoxic potential.
