In Vitro investigation of T-PRF membrane structure containing different doses of doxycycline

Abstract (EN)

The aim of this clinical study was to inject 0.5 ml, 1 ml and 2 ml Doxycycline into T-PRF membranes obtained from blood collected from patients and to examine the fibrin network pattern, antimicrobial activity, drug release, degradation properties of the membranes by light microscopy and to compare them with the control group. Our study was performed on a total of 15 healthy individuals without any bleeding disorder. The obtained T-PRF membranes were evaluated according to the amount of degradation by degradation, antibacterial activity against S. aureus and P. aeruginosa bacteria, time-dependent drug release concentration and fibrin network pattern parameters by light microscopy. According to the results of statistical analysis, 0.5 ml, 1 ml, 2 ml drug-loaded T-PRF membranes showed significantly lower degradation percentage compared to non-drug loaded T-PRF membranes (p≤0.001). When the fibrin network pattern scores of drug loaded and non-drug loaded membranes were compared, the fibrin network pattern scores of drug loaded T-PRF membranes were significantly higher (p≤0.001). Antibacterial activity examination revealed no antibacterial activity against P. aureginosa in all groups. When antibacterial activity against S. aureus was examined, higher inhibition zone diameter was observed in the 2 ml drug loaded T-PRF membrane group. 2 ml drug-loaded T-PRF membranes did not show higher inhibition area diameter than other drug-loaded T-PRF membranes.
