Abstract (EN)

An important factor that increases the success of artificial insemination is that veterinarians who perform artificial insemination practice under field conditions take into account the appropriate conditions while thawing frozen semen straws. Regarding its effect on the survival of the spermatozoa, the thawing procedure is just as significant as the freezing procedure. In this study, two alternative thawing temperatures will be compared and evaluated for their usability, validity, and accuracy in frozen and thawed semen exams. Additionally, it is to statistically compare and contrast the results from spermatological parameters with the semen parameters waiting at various intervals after thawing. In the study, 20 straws of frozen semen from the same bull, frozen on the same date and stored under the same conditions, were used. Two different groups were formed for the study design. First group, 10 straws of semen samples were thawed in a 37°C water bath for 30 seconds, and second group, 10 straws of semen samples were thawed at 70°C for 5-6 seconds. All thawed semen samples were left at +4°C, and spermatological examinations were performed at 0, 3, 6, 12, 24, and 48 hours. In this study, as a result of spermatological examination of semen thawed at different thawing temperatures of 37°C and 70°C, respectively, motility, viability rate, and sperm morphology are evaluated, and it has been observed that waiting times can cause significant differences. It was determined that as the waiting time increased after thawing, total motility in semen decreased. At the same time, it was revealed that sperm morphology was negatively affected as the waiting time increased after the semen thawing. This study demonstrated the necessity of doing more in vitro and in vivo studies to increase the post-thaw efficiency of semen used in artificial insemination applications.
