Abstract (EN)
The purpose of this thesis is to investigate the possibilties of mass propagation of Foşa, Tombul and Çakıldak which are the most important hazelnut varieties in Turkey using different PGRs. For shoot development DKW basic nutrient medium including different doses of PGRs like BAP (0, 1, 2, 3, 4, 5 mg/L), IBA (0, 0.01, 0.1, 0.2 mg/L), GA3 (0.05 mg/L) and MT (0, 1, 2, 3, 4, 5 mg/L) were tested. The best shoot development was obtained in the media including 2 mg/L MT, 0.2 mg/L IBA and 0.05 mg/L GA3 for Foşa and Tombul and for Çakıldak the media including 3 mg/L MT, 0.2 mg/L IBA and 0.05 mg/L GA3. For rooting MS basic nutrient medium including different doses of PGRs like IBA (0, 1, 2, 3 mg/L), NAA (0, 0.01, 1, 2 mg/L) and 200 mg/L Sequestren Fe 138 were tested. The best root obtained the media including 2 mg/L IBA and 1 mg/L NAA for Foşa, Çakıldak and Tombul. Thermotherapy was applied to the plants infected with ApMv in vitro for elimination of this virus. During thermotherapy, these plants were kept in 16 h light/8 h dark photperiod, 40 μmol∙m2∙s-1 light intensity with 25˚C and day by day temperature increasing 1˚C reached to 36˚C in light and 32˚C in dark at 28-32 weeks. In thermotherapy application, these plantlets were tested with DAS-ELISA and RT-PCR and the virus in these plants were eliminated with thermotherapy application in 45th days.
