Fosfat çözücü bakterinin biyoenformatik analizlerle genom haritasının çıkarılması

Pseudomonas species is known as a plant growth-promoting bacteria as well as a good phosphate solubilizer. Pseudomonas species is commonly used in the studies, not only in the biotechnological and agricultural fields, but also in the field of medicine. In this study, the whole genome sequencing of the Pseudomonas species isolated from the root of the IZA wheat (Triticum monococcum spp. monococcum) in Seben/Bolu was performed using the next generation sequencing (Illumina NextSeq 550) with a total of 2,188,517 paired-end reads, were totally 655,555,100 bases, in 76x depth. Paired-end reads were assembled using de novo assembly approach and was created the draft genome of Pseudomonas spp. ESU1531 with a 252 contig with a GC content of 61.03% and a length of 6,520,191. Annotation of all the genes in the genome was done by PROKKA. According to the PROKKA result, the coding genes in the genome was predicted as 5652 and 4547 of them were functionally assigned. 1 rRNA and 66 tRNA were found in the genome. In addition, phylogenetic of 16s rRNA and multi-locus analyses were done and Pseudomonas spp. ESU1531 taxonomically classified. The whole genome sequencing and analysis of Pseudomonas spp. ESU1531 will benefit many different studies. In particular, it will benefit studies for the use of bio fertilizers and/or biocontrol agents in the biotechnological and agricultural fields; on the other hand, it will contribute to future studies in understanding the phosphate solubilizing mechanism of the Pseudomonas species. Moreover, Pseudomonas spp. ESU1531 will also be a very important reference in genomic and/or metagenomic studies in the Seben region in the future.
