The effect of rolipram, a fosfodiesterase 4 inhibiting agent, on kidney reperfusion injury

Purpose: Renal ischemia is a common entity that seen in renal transplantation, partial nephrectomy, sepsis, and a variety of urological procedures and various clinical situations, such as hydronephrosis. Therefore, the importance of nephron-sparing surgery is increasing with time. To minimize the damage of renal ischemia in the kidney tissue, various pharmacological agents studied experimentally. These agents include the various phosphodiesterase inhibitors. In this study, we aimed to investigate the effect of the Rolipram as agent of phosphodiesterase 4 inhibitor, on renal reperfusion injury.Method and Materials: The study was planned by separation of 30 rats to 5 different groups containing 6 in each. In the first group nothing was done. In the second group the renal pedicle clamped for 30 minutes. In the third group, 30 min before clamping, in fourth group when clamp is placed, in fifth group after 30 minutes opening the clamp, 1mg/kg rolipram was given intraperitoneally by injection. Clamping time was determined as 30 minutes. Nephrectomy was performed 24 hours after all of these groups. For each kidney, half of the tissues examined histopathologically for inflammation, tubular necrosis and apoptosize views. The other half of the kidneys examined by biochemical methods including malondialdehyde, superoxide dismutase, catalase levels measuring.Findings: In rats with the administration of Rolipram, renal I / R injury significantly decreased. In oxidative damage, malondialdehyde levels, which is stable metabolite of lipid peroxidation caused by free radicals was significantly higher in second (I / R) group than fifth group in particular. At the same time that the antioxidant enzyme superoxide dismutase and catalase levels were significantly higher in Group 2 than group 5. Histopathologically, the inflammation of the groups which administrated Rolipram compared with group 1 (Control) and 2 (I / R), there was not a very significant difference. Tubular necrosis and apoptosis in group 5 was significantly lower than the other groups except group1(control).Result: We consider that Rolipram is an opinion in surgical procedures which can lead to renal I / R injury and can decrase oxidative renal tissue damage and severe deterioration of renal functionKey Words: Rolipram, renal ishemia, ishemia reperfusion injury ( İ / R )
