Investigation of the effect of lactobacillus plantarumon cardiac AKT/enos pathway in fructose-induced metabolic syndrome

Abstract (EN)

Aim: It has been shown that fructose, which is widely used in the diet, increases the risk of developing metabolic syndrome. Metabolic syndrome, on the other hand, increases the risk of cardiovascular disease in relation to oxidative stress and inflammatory variables. Lactobacillus plantarum (L. plantarum), one of the lactic acid bacteria, has antioxidant, anticancer, anti-inflammatory, antiproliferative, anti-obesity and antidiabetic effects. There are a limited number of studies in the literature examining the effects of probiotic strains of lactic acid bacteria on cardiovascular diseases in models with fructose-induced metabolic syndrome. In this study, it was investigated whether L. plantarum supplementation had an effect on the Akt/eNOS pathway in the hearts of fructose fed rats. Material and Method: This study was conducted in Wistar Albino male rats with fructose-induced metabolic syndrome. Rats control, fructose and fructose+L. plantarum were divided into three groups. While fructose is given as a 20% solution in drinking water for 15 weeks; probiotics were administered by gastric gavage once a day (109CFU/ml/100 g) in the last 6 weeks of fructose administration. Akt, eNOS, iNOS, IRS-1, SIRT1, mTOR, NF-kB and TNF-α protein levels in heart tissues were evaluated by Western blot analysis methods. Results: According to the fructose group of L. plantarum application, Akt level was 24.7% (p<0.0001), IRS-1 level was 43.7% (p=0.0113), eNOS level was 25.7% (p<0.0001), mTOR level was 35.1% (p<0.0001) in heart tissue of rats compared to fructose group. ) increased SIRT 1 level by 41.7% (p=0.0031) and increased iNOS level by 25.0% (p<0.0001), NF-kB level by 27.6% (p<0.0001), and TNF-α level by 26.1% (p<0.0001) found to be reduced. Conclusion: As a result of the study, it has been shown that L. plantarum can prevent heart inflammation, contribute to increasing insulin sensitivity through Akt/eNOS pathway, and prevent cardiovascular disorders that may occur due to metabolic syndrome.
