Abstract (EN)

Despite the advanced examination and treatment opportunities; they continue to be significant today due to the fact that gastrointestinal system (GIS) carcinomas are among the most mortal malign tumors. Depth of invasion and metastasis are the most important prognostic factors in GIS carcinomas. In addition to these, many different independent prognostic factors such as serum CEA level, microscopic type and bcl-2 expression have been identified. In spite of such many prognostic factors determined, clinical and molecular heterogeneity observed in colorectal cancers requires the search for new prognostic factors.The overexpression of EZH2, a receptor of gene transcription (enhancer of zeste homolog) is associated with biological malignancy in many malignant tumors. It has been observed that E-cadherin, an adhesion molecule in class of tumor suppressor gene, has negative correlation with EZH2 in prostate carcinoma. In addition, ıt is argued that the EZH2 is associated with other genes such as p53 and Ki67 in tumor development.In this study, we aimed to research relationship of EZH2 expression with E-cadherin, Ki67 and p53 expressions and clinicopathologic parameters in GIS carcinomas, and to reveal whether or not EZH2 can be used as prognostic marker in the GIS carcinomas.A total of 90 cases of gastric and colorectal resection materials, comprising 60 cases with colorectal resection and 30 cases with gastric resection were included in this study. Clinicopathologic parameters such as metastasis, stage and grade for each case were determined. Parafin sections were obtained and stained with EZH2, p53, Ki67 and E-cadherin immunohistochemically.When examined the relationship of EZH2 staining extensiveness and intensity with clinicopathologic parameters in gastric and colorectal carcinomas, it has been observed that both extensiveness and intensity have increased in advanced stage, metastatic and poorly differentiated cases. When examined the relation of the EZH2 with other immunohistochemical markers, there was positive correlation between EZH2 and p53, and between EZH2 and Ki67. The correlation between EZH2 and E-cadherin was negative but extremely weak. In our study, the staining extensiveness and intensity of the EZH2, p53, Ki67 and E-cadherin were also exemined in metastatic tumor tissues and main tumor tissues. It appears that the staining extensiveness and intensity of E-cadherin was higher in the main tumor tissues in both gastric and colorectal carcinomas. The staining extensiveness and intensity of the EZH2, p53 and Ki67 was higer in metastatic tumor tissues.At the end of the study, when considered relationship of EZH2 with clinicopathologic parameters and other immunohistochemical markers, it has been observed that this marker can be used in determination of the prognosis. At the same time, EZH2 can give us an opinion about the degree of tumor progression together with other immunohistochemical markers.
