The investigation of grapevine yellow speckle disease (gysvd-1 and 2) in Gaziantep grapevine areas

This study was conducted to determine presence, infection rates and characterization of factors for Grapevine yellow speckle (GYS) disease of Gaziantep vineyards. Gaziantep region GYS disease was examined approximately in 4500 da on the 766363 grapevine plants for macroscopic symptoms of yellow speckle and vein banding. 652 grapevine plants were found to be infected with grapevine yellow speckle disease. The infection rate is % 0,08 in Gaziantep vineyards. At the 67 grapevine plants in infection, the viroid bands of GYSVd-1 and 2 were observed in sPAGE analysis. The infection rate of GYS disease at organic grapevine yards was found to be % 0,04. In organic varieties of Dımışkı and Öküzgözü, viroid bands of GYSVd-1 and 2 were also found as a result of sPAGE analysis that were observed on acrylamide gel. GYS disease was found at the % 0,49 infection rate on grapevine seedlings at Pistachio Research Institute. In the Institute the mother blocks were determined to be clean about GYS disease. RT-PCR studies GYSVd-1 and 2 were determined on agarose gel. Nucleotide analysis of the length of GYSVd-1 was found as 366 nts. However the results of GYSVd-2 was to be 361 nucleotides.
