Cancer is a disease caused from the changes in the critical genes that control cell proliferation, differentiation, survival. Cancer and apoptosis are often seen in the cell population dynamics as mutually case. Uncontrolled cell proliferation and decreased apoptosis contribute to the development of colon cancer. Geldanamycin is a benzoquinone antibiotic ansamisin that showing the anti-cancer effect. HSPs are molecular chaperones that synthesized in response to heat stress, regulate cellular homeostasis and improve overall survival of the cell. HSP90 is heat shock protein that impeding the realization of programmed cell death in cells due to the its protective nature. In our study we aimed to examine the effects of 17-AAG, on Colo?205 colon cancer cell line index to investigate culture on apoptosis, primarily the method of MTT viability and growth characteristics of the cells, and then the TUNEL method to identify apoptotic cell ratio. Then the effect of 17-AAG on apoptosis on the signal path, the path key mitochondrial proteins, cytochrome c, bcl?2, caspase?9, apaf?1 expression in RT PCR method were evaluated. After the implementation of 17-AAG, with MTT analysis, cell proliferation stopped and death occurred. When RT-PCR is analyzed, gene expression in cytochrome c, apaf?1, caspase?9 increased in a statistical manner (p <0.05) and Bcl?2 expression significantly decreased in a remarkable way (p <0.05). Therefore, 17AAG application seems to be an agent that may contribute to the treatment of colon cancer. More studies on this topic would be useful for supporting these findings.
