Abstract (EN)
Aim: The aim of this study is to investigate the role of the "Pre‑mir‑27a variant rs895819" gene polymorphism in Gestational Diabetes. In the previous studies it was found that there is a relationship between this gene and Type 2 diabetes but there is no study on gestational diabetes. Thus, this study investigating the relation between the gene polymorphism Premir27a variant rs895819 and GDM is the first one conducted on this issue. Material and Method: Non-kin 106 patients (working group) diagnosed Gestational Diabetes in the Department of Obstetrics and Gynaecology of the Medical Faculty in Zonguldak Bülent Ecevit University and 100 healthy pregnant women with non chronic disease (control group) took part in this study. After the information and consent period, in order to investigate only related gene polymorphism DNA isolation of 2 ml of donated blood sample was handled via kit method for each individual's routine control. The suitability of DNA quality was determined by measuring the obtained Genomic DNA at spectrophotometer in 280 nm wave length. The Pre‑mir‑27a variant rs895819 gene polymorphisms were conducted through PCR_RFLP method with appropriate primers. For the analysis of the study, SPSS 18.0 for Windows (Chicago, IL) packaged software was used. Findings and Results: A significant difference between two groups was determined in terms of TT, TC and CC genotypes in the statistical analysis performed on 106 patient group and 100 control group. Other analyses were also performed on phenotype according to the models in which C allele is dominant or recessive, but there is no significant difference.
