Effect on prognosis and survival on treatment results of glioblastoma multiforme patients treated with concomitant and adjuvant temozolomide

Abstract (EN)

Purpose: Glioblastoma multiforme (GBM) in adults is the most common malignant primary brain tumors. GBM after treatment most common problems in follow-up, maintain local control. In this study, GBM treatment kemoteropatic used temozolomide agent in December 2005 in our country temozolomide be licensed after the protocol in accordance with radiotherapy after surgery and chemotherapy in the treatment of 28 cases results, before this date after surgery, we provide only radiotherapy, randomly selected 26 cases temozolomide results 'in order to evaluate the effectiveness and safety were evaluated.Methods and Materials: Pathologically confirmed, according to the protocol features, glioblastoma multiforme diagnosed cases received were divided into two groups. The first group treated with radiotherapy after surgery only cases, the second group (combined treatment group) treated with postoperative radiotherapy and temozolomide as patients were taking. All cases local fractional irradiation (60 Gy total dose: 2 Gyx5 days / week, for 6 weeks) were performed. Just as the second group concomitant orally 75 mg/m2/günx7gün/hafta, as monotherapy for 6 weeks followed by 200 mg/m2/günx5gün, six cycles every 28 days, underwent a temozolomide. In addition Temozolomide?s to effects on prognosis and survival age, gender and size of resection of the median in both groups until the last time (PSS), and median overall survival time (GSS) The effects were evaluated independently.Results: Work as a result of PSS, combined therapy group, 14 months, radiotherapy alone group, only 6 months, respectively (P <0.0001). GSS, the combined treatment group, 16 months, radiotherapy alone in the group is 12.5 months, respectively (P = 0.0354). Both grups important prognostic factors, age and sex for both PSS, as well as non-statistically significant benefit in terms of GSS are being identified. For a resection size (total-subtotal), statistically significant benefit was found.Conclusion: Total combined post-surgical treatment (RT + TMZ), prognosis and survival compared to radiotherapy alone has been shown to be more effective on.Key Words: Glioblastoma multiforme, Temozolomide, Treatment
