Abstract (EN)

A total of 120 swab samples were taken from both healthy (n=60) and unhealthy (n=60) individuals of these different ages and both genders. Vitek II technology, traditional biochemical and culture tests were used for the diagnosis of bacterial isolates. In samples taken from sick individuals, 47% of the isolates were found to belong to Streptococcus mutans. The susceptibility of bacterial isolates to antibiotics was determined by the Kirby-Bauer disk diffusion technique and most of the isolates showed resistance to antibiotics. Most of the S. mutans isolates produced biofilms by the microplate method. Detection of 16s rRNA and gtf gene in isolates was demonstrated at a statistically significant rate using polymerase chain reaction. Locally isolated Lactobacillus sakei was screened to select isolate active in bacteriocin production. The antibacterial activity of bacteriocin was high on all 10 S. mutans isolates with respect to zones of inhibition ranging from 19 to 23 mm at 200 µg/mL. In this thesis, we aimed to determine the gtf gene molecularly and to examine the relationship between bacteriocin production and oral flora and pathogenic bacteria that cause oral disease.
