Evaluation of cytotoxic and genotoxic effects of silver tin alloy nanoparticles on healthy lung cell line

Abstract (EN)

In this study, cytotoxic and genotoxic effects of Ag-Sn metal alloy nanoparticles in BEAS-2B human lung epithelial cells were evaluated. XXT test was used to determine cytotoxic effects, Comet test was used to determine genotoxic effects. ROS test was applied to determine the presence of reactive oxygen species. Before starting the experiments, zeta potential, DLS and TEM analyzes were used to characterize the nanoparticles. For XXT test, BEAS-2B cells were exposed to different concentrations (1 to 1000 ml) of Ag- Sn alloy nanoparticles for 24, 48 and 72 hours. To determine the genotoxic effect of the Ag-Sn alloy nanoparticle, BEAS-2B cells were treated with nanoparticles different concentrations ( 50 to 500 ml) for 24, 48 and 72 hours and a Comet method was applied. According to the results obtained, it was observed that the cytotoxic effect of Ag-Sn nanoparticle increased different concentration and exposure time. Comet test showed that exposure to Cu-Zn alloy nanoparticles induced single and double stranded DNA damage in BEAS-2B cells. Our results further indicated that increased intracellular ROS formation could be related to the experiments that these effects.
