Abstract (EN)
In this thesis study, it was aimed to propagate the plant of Gypsophila simonii HUB.-MOR. using in vitro techniques. This plant is a local endemic in Çankırı Province with arid and semi-arid lands, and an endangered (EN) species taking part among medicinal and aromatic plants. Therefore, 3 different basal media were used for in vitro propagation of G. simonii [a) Murashige and Skoog b) Nitsch & Nitsch c) Gamborg's B5], a solidifier (2.1 g/ L gelrite), 6 cytokinins and 6 auxins [a) 6-benzylaminopurine (BAP) (0 mg/L, 0.5 mg/L, 1 mg/L), b) Kinetin (KIN) (0 mg/L, 0.5 mg/L, 1 mg/L) c) Indole3-butyric acid (IBA) (0 mg/L, 0.25 mg/L, 0.5 mg/L), d) α-naphthalene acetic acid (NAA) (0 mg/L, 0.25 mg/L, 0.5 mg/L)] along with plant growth regulators prepared in 36 different combinations with 30 g/L sucrose was added to the basal media and as a carbon source, and the pH of the medium was set 5.7. During the in vitro propagation of the plant, superficial and internal infections were frequently encountered, so the experiment was repeated twice and this situation was resolved with the protocol developed for the plant. The highest shoot number (40) was obtained in MS nutrient medium containing the combination of KIN (0 mg/L) + NAA (0.5 mg/L) PGR. In the rooting stage, MS basal medium was more effective. The best callus development is in MS basal medium, 0 mg/L BAP + 0.5 mg/L IBA, 0.5 mg/L BAP + 0 mg/L IBA, 0.5 mg/L BAP + 0.5 mg/L IBA, 0 mg/L BAP + 0.25 mg/L NAA 0 mg/L BAP + 0.5 mg/L NAA, 0.5 mg/L BAP + 0.5 mg/L NAA, 0 mg/L KIN + 0.25 mg/L IBA, 0 mg/L KIN + 0.5 mg/L NAA, supplemented with plant growth regulator. It is important that in vitro reproduction studies of this endemic species have not been carried out sufficiently and that it will form the basis for future studies.
